Construction of a Quinoline-based Sequential Functioning Chromogenic Sensor for Copper(II) Ion and Biothiols: Its Application to Test Strips

A new compound BQ ((1E,1’E)-N,N’-((1,3-phenylenebis(oxy))bis(3,1-phenylene))bis(1-(quinolin-2-yl)methanimine)) containing quinoline moiety has been developed as a sequential functioning chromogenic sensor for copper(II) ion and biological thiols (GSH, Cys, and Hcy). BQ had a clear chromogenic response toward only copper ion with a red-shifted absorption. Detection limit for copper(II) ion was calculated to be 1.20 mu M. Its application was successfully conducted in test strips. The resultant BQ-Cu2+ complex could detect the biological thiols, showing the color change and blue-shifted absorption via the demetallation of copper(II) ion. The possible binding mechanisms of BQ to copper(II) ion and of BQ-Cu2+ to biological thiols were understood by UV-vis titrations, Job plot, ESI-MS, and H-1 NMR titrations.

Binary and ternary copper(II) complexes with 6-ethyl-4-hydroxy-2,5-dioxo-5,6-dihydro-2H-pyrano[3,2-c]quinoline-3-carboxaldehyde: Synthesis, spectral, thermal, molecular modeling, antimicrobial and antitumor studies

6-Ethyl-4-hydroxy-2,5-dioxo-5,6-dihydro-2H-pyrano[3,2-c]quinoline-3-carboxaldehyde (HL) was synthesized as a ligand and subject to chelate with Cu(II) ion of different anions (AcO-, NO3-, SO42-ClO4-, Cl- and Br-) in presence and absence of auxiliary ligands (L’); N,O-donor; or N,N-donor such as 8hydroxyquinoline, 1,10-phenanthroline and 2,2′-bipyridyl. The obtained results revealed that the ligand behaves as a neutral/monoanionic bidentate, forming chelates with molar ratio 1:1, (M:L). The metal complexes were fully characterized by analytical and spectral techniques in addition to thermal, magnetic susceptibility and conductivity measurements. The metal complexes displayed square planar and octahedral geometrical arrangements depending on the nature of the anion. The ligand and its complexes were screened for their antimicrobial activity against Gram-positive bacteria which are Staphylococcus aureus and Bacillus subtilis, Gram-negative bacteria which are E. coli and S. typhimurium, yeast (Candida albicans) and fungus (Aspergillus fumigatus). The antitumor activity of the HL ligand and its Cu(II) complexes were examined against Ehrlich Ascites Carcinoma cell line (EAC) from ascetic fluid of the female Swiss albino mice. To explore the most probable structure and the nature of bonding of HL ligand to metal ions; semiemperical (PM3) calculations were achieved in gas phase. 8-Hydroxyquinoline platinum(ii) loaded nanostructured lipid carriers: synthesis, physicochemical characterization and evaluation of antitumor activity

Every year the incidence of cancer and the death rate are increasing worldwide. The drug 8-hydroxyquinoline platinum(ii) [PtCl(8-O-quinoline)(dmso)] (8HQ-Pt) has been identified as a promising antitumor complex. Nanostructured lipid carriers (NLC) are second-generation drug nanocarrier systems that have superior advantages over other kinds of colloidal carrier systems. 8HQ-Pt compound loaded NLC formulations of cetyl esters (SS) were synthesized via ultrasonication in the presence of two different liquid oils: capric triglyceride, or olive oil. The physicochemical and microscopic characterizations of NLC were analyzed via dynamic light scattering (DLS), transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FTIR), thermogravimetric analysis (TGA), differential scanning calorimetry (DSC) and X-ray diffraction analysis (XRD). In vitro drug release and cytotoxicity, cell uptake and apoptosis assays against the human colon cancer cell line HT-29 were investigated. The results showed that NLCs indicated a narrow size distribution and a mean particle diameter in the range of 136-159 nm. The thermal characteristic analysis confirmed the stability of NLCs up to 185 degrees C. Encapsulation efficiencies of the 8HQ-Pt compound in NLCs were about 80% and the 8HQ-Pt compound in the formulations showed a controlled release profile during 72 h. The release profiles of these two different formulations and the antitumor effect on the HT-29 cell line were compared with those of the free 8HQ-Pt compound. The cellular uptake of two different NLC groups was proved by fluorescence microscopy and the presence of capric triglyceride liquid oil in the formulation increased the intracellular drug delivery capacity when compared with olive oil.

An efficient silver-catalyzed d-regioselective C(sp(3))-H heteroarylation of free alcohols has been developed. A novel chemosensor for the distinguishable detections of Cu2+ and Hg2+ by off-on fluorescence and ratiometric UV-visible absorption

A novel dual-functional chemosensor, derived from the conjugation of rhodamine B with a quinoline derivative (RHQ), was firstly synthesized with high efficiency and cost-effectiveness for the distinguishable detections of Cu2+ and Hg2+ via ring-opening and ring-forming mechanism. The chemosensor exhibits highly selective and distinguishable responses for Cu2+ and Hg2+ in CH3CN-H2O (4:1, v/v) with off-on fluorescence and ratiometric ultraviolet-visible (UV-Vis) absorption changes. Additionally, Cu2+ is identified by opening a rhodamine spirocycle with a UV-Vis absorption band, at around 560 nm and fluorescence turn-on. Interestingly, Hg2+ is discerned by opening the rhodamine spirocycle and by generating a new special cycle for the quinoline unit. Resultantly, there were two UV-Vis absorption bands at around 365 nm and 560 nm, which were accompanied by fluorescence turn-on. Moreover, the chemosensor can quantitatively detect Cu2+ and Hg2+ by off-on fluorescence and ratiometric UV-Vis absorption changes, respectively. Furthermore, the chemosensor with low cytotoxicity could be successfully administered to monitor Cu2+ and Hg2+ in living cells. This work may pay the way for the development of dualfunctional chemosensor for quantificationally detecting metal ions in environmental and biological systems. (C) 2020 Elsevier B.V.
