Novel dyes based on the 12H-quinoxalino[2,3-b] [1,4]benzothiazine skeleton were synthesized and subsequently characterized using 1H NMR. Their electrochemical and spectral properties, such as absorption and emission spectra, quantum yield of fluorescence, and quantum yield of singlet oxygen formation, were measured. These compounds were evaluated as sensitizers for alkoxypyridinium salt photodecomposition, and the results are discussed on the basis of the free energy change for electron transfer from benzothiazine dyes to alkoxypyridinium compounds. Benzothiazine dyes are useful oxidizable sensitizers for N-alkoxypyridinium photoinitiators. The mechanism of the dye photobleaching is supported by time-dependent density functional theory (TD-DFT) calculations and the quantum yields of sensitized proton formation. Photoredox pairs consisting of benzothiazine dyes and alkoxypyridinium salt were found to be effective initiation systems for free radical polymerization of methyl acrylate and trimethylolpropane triacrylate (TMPTA) using visible light.

The synthesis of oxacalix[2]terphenylene[2]aromatics with enlarged macrocyclic holes by cyclooligomerization reaction of 5?-tert-butyl-(1, 1?:3?,1?-terphenyl)-3,3?-diol 1 with electron-deficient dihalogenated benzene and azaheterocycles is described. The structures of the macrocycles were characterized by NMR, HRMS spectroscopic and X-ray diffraction techniques. Single crystal X-ray analysis revealed that the terphenyl-3, 3?-dioxo unit incorporated in the oxacalix[4]aromatics scaffold can adopt all three possible conformations (I, II, III). The cis-conformational terphenyl-3,3?-dioxo (I and II) derived oxacalix[4]aromatics were found to adopt both chair and boat conformations, resulted in creation of molecular cavities capable of hosting solvent molecules of chloroform. The trans-conformational terphenyl-3,3?-dioxo (III) derived oxacalix[4]aromatics, however, adopt a twisted chair conformation with a narrow void space incapable of hosting any guest molecules.

We report the successful synthesis of a series of ethynylated dioxadiazaacenes and investigate their properties. We developed a modular Cu-based catalytic procedure to build up [1,4]dioxino[2,3-b]pyrazine motifs starting from only a few building blocks. TIPS-acetylene-substituted benzene-1,2-diol and naphthalene-2,3-diol were reacted with 2,3- dichloropyrazine, 2,3-dichloroquinoxaline, and 2,3-dichlorobenzoquinoxaline to give a set of six novel and well-soluble dioxadiazaacenes. Different reaction conditions for the coupling were tested. Copper catalysis is most effective and gave the best yield of dioxadiazaacenes. The resulting azaoxaacenes were characterized in terms of optical and electronic properties and crystal packing.

A new class of PDE4 inhibitors were designed and synthesized via the InCl3 mediated heteroarylation of indoles and their further derivatization through the Pd(II)-catalyzed C[sbnd]H activation strategy. This effort allowed us to discover a series of 2-(1H-indol-3-yl)-quinoxaline based inhibitors possessing PDE4B selectivity over PDE4D and PDE4C. One of these compounds i.e. 3b (PDE4B IC50 = 0.39 ± 0.13 muM with ?27 and > 250 fold selectivity for PDE4B over PDE4D and C, respectively)showed effects in Zebrafish experimental autoimmune encephalomyelitis (EAE)model of multiple sclerosis when dosed at 3, 10 and 30 mg/kg intraperitoneally. Indeed, it halted the progression of the disease across all these doses tested. At an intraperitoneal dose of 30 mg/kg the compound 3b showed promising effects in adjuvant induced arthritic rats. The compound reduced paw volume, inflammation and pannus formation (in the knee joints)as well as pro-inflammatory gene expression/mRNA levels significantly in arthritic rats. Moreover, this compound was found to be selective towards PDE4 over other families of PDEs in vitro and safe when tested for its probable toxicity (e.g. teratogenicity, hepatotoxicity and cardiotoxicity)in Zebrafish.
