Bronsted acid catalyzed alkylation of indoles with tertiary propargylic alcohols: Scope and limitations

Direct alkylation of indoles with a wide variety of tertiary propargylic alcohols under Bronsted acid catalysis conditions has been studied. A general and environmentally friendly method for the synthesis of 3-propargylated indoles with quaternary carbon atoms at their propargylic positions has been developed. The reactions are highly regioselective with regard both to the indole and to the alkynol components. Only with N-unsubstituted 2-arylindoles do competitive SN? reactions take place to afford 3-dienyl- or 3-allenylindoles, depending on the alkynol moiety. The reactions were carried out in air with undried solvents, and water was the only side product. Treatment of indoles with tertiary propargylic alcohols in the presence of Bronsted acid catalysts provided 3-propargylindoles with quaternary centers at their propargylic positions. In addition, 3-dienyl- and 3-allenylindoles could also be obtained with convenient substitution in the indole and alkynol components. A simple method for the synthesis of various 3-(4,5-dihydro-1H-imidazole-2-yl)-1H-indoles is described. Treatment of different substituted indoles with 1-acetyl-imidazolidin-2-one in the presence of phosphorus oxychloride afforded after hydrolysis in ethanol the corresponding 3-(4,5-dihydro-1H-imidazole-2-yl)-1H-indoles in moderate to good yields.

A highly active catalyst for the reductive cyclization of ortho-nitrostyrenes under mild conditions

A mild and efficient method for the palladium-catalyzed reductive cyclization of ortho-nitrostyrenes to afford indoles is reported. Treatment of ortho-nitrostyrenes with 0.1 mol% palladium (II) trifluoroacetate [Pd(TFA) 2] and 0.7 mol% 3,4,7,8-tetramethyl-1,10-phenanthroline (tm-phen) in DMF at 15 psig CO and 80C afforded indoles in good to excellent yields. When the reaction was conducted in toluene, the corresponding N-hydroxyindole was isolated. A mechanism that accounts for the formation of N-hydroxyindole is proposed.

An efficient synthesis of 6H-isoindolo[2,1-a]indol-6-ones through rhodium-catalyzed NH-indole-directed C-H carbonylation of 2-arylindoles with carbon monoxide has been developed. Preliminary mechanistic studies revealed that this reaction proceeds via N-H bond cleavage and subsequent C-H bond cleavage. Reaction monitoring via ESI-MS was used to support the formation of five-membered rhodacycle species in the catalytic cycle.

Iron-catalyzed reductive coupling of nitroarenes with olefins: Intermediate of iron-nitroso complex

Using a single half-sandwich iron(II) compound, CpFe(1,2-Ph2PC6H4S)(NCMe) (Cp- = C5Me5-, 1) as a catalyst, reductive coupling of nitroarenes with olefins has been achieved by a well-defined iron(II)/(EtO)3SiH system. Through either inter- or intramolecular reductive coupling, various branched amines and indole derivatives have been directly synthesized in one-pot. Mechanistic studies showed that the catalysis is initiated by activation of nitroarenes by the iron(II) catalyst with silane, generating iron-nitrosoarene intermediate for the C-N bond coupling.
