Ligand/PTC-free intramolecular Heck reaction: Synthesis of pyrroloquinoxalines and their evaluation against PDE4/luciferase/oral cancer cell growth in vitro and zebrafish in vivo

A series of 1,3-disubstituted pyrrolo[2,3-b]quinoxalines has been designed for the potential inhibition of PDE4 without inhibiting luciferase. A ligand/PTC (phase transfer catalyst) free intramolecular Heck cyclization strategy was used to prepare these compounds, some of which showed significant inhibition of PDE4B (IC50 ? 5-14 muM) and growth inhibition of oral cancer cells (CAL 27) but not inhibition of luciferase in vitro. They also showed acceptable safety profiles but no apoptosis in zebrafish embryos.

Synthesis of novel oxidizable polymerization sensitizers based on the dithiinoquinoxaline skeleton

Novel dyes, based on the dithiinoquinoxaline skeleton, were synthesized and characterized using 1H NMR spectroscopy and chemical ionization mass spectroscopy. Their spectral properties, such as absorption, emission spectra and quantum yield of fluorescence, were also measured. Electron donating properties of the title compounds were estimated on the basis of DFT calculations. The studied dyes were used as oxidizable sensitizers for 2,4,6-tris(trichloromethyl)-1,3,5-triazine (Tz). The dye/Tz photoredox pairs were found to be effective visible-wavelength initiators of free radical polymerization. The ability of these systems to act as photoinitiators strongly depended upon the free energy change of the photoinduced electron transfer from the excited dyes to Tz. It has been shown that the intermolecular electron transfer is the limiting step in the photopolymerization initiated by these studied initiator systems.

Cyclization Reactions through the SRN1 Mechanism. Reactions of o-Dihaloaromatic Compounds with Dithiolate Ions

In the present paper we report the reactions of o-dihaloaromatic compounds with dithiolate ions as new cyclization reactions occuring by the SRN1 mechanism. o-Diiodo- and o-bromochlorobenzene react under photostimulation with 3,4-toluenedithiolate ion to give good yields (ca. 60percent) of the cyclic disubstituted compound 2-methylthianthrene.The reaction occurs to a lesser extent (ca. 20percent) when the substrate was 1-bromo-2-iodonaphthalene. 2,3-Dichloroquinoxaline reacts with the same nucleophile in the dark or in the presence of radical traps to give a 100percent yield of the cyclic disubstituted compound.The reaction of o-diiodobenzene with 1,2-ethanedithiolate gave a low yield of the cyclic compound benzo-1,4-dithiane.This decrease was explained in terms of fragmentation reactions of the proposed radical anions intermediates.

Studies in the syntheses of s-triazolo[4,3-a] quinoxalines

o-Phenylendiamine 1 is condensed with oxalic acid using Phillip’s procedure to obtain quinoxaline-2,3-doine 2, which on treatment with POCl3 gives the known 2,3-dichloroquinoxaline 3. 2-Chloro-3-hydrazinoquinoxaline 4 is prepared in a facile and simple way by treatment of 3 with hydrazine hydrate in methanol or in dioxane containing triethylamine. Condensation of 4 with a variety of aldehydes in DMF at room temperature furnishes the corresponding arylidene/alkylidine derivatives 5 which undergo smooth nucleophilic substitutions of chlorine by alkoxide or phenoxide ions yielding 2-alkoxy/phenoxy-3-(2-arylidene/alkylidene hydrazino) quinoxalines 6. The dehydrogenative cyclisation of 6 is achieved with chloranil in refluxing 1,2- dichloroethane resulting in s-triazole-[4,3-a] quinoxalines 7 whose structures are supported by spectral and analytical data and by alternate chemical synthesis in the case of the parent compound 7a.

Fused pyrazino[2,3-b]indolizine and indolizino[2,3-b]quinoxaline derivatives; synthesis, structures, and properties

The synthesis of six new compounds incorporating either a pyrazino[2,3-b]indolizine or indolizino[2,3-b]quinoxaline core are reported in good yield (58-87%). The intermediates for the key cyclization reaction for one set of compounds (5a-c), with a sterically demanding 3,5-dimethylpyrazole group in the 5-position of the core, were found to be mono-substituted. These intermediates could be isolated and cyclized by heating under acid-catalyzed conditions. To further demonstrate the versatility of the chemistry, compounds 6a-c were synthesized in 58-68% yields. Compounds 5a-c are non-planar in solution and the solid-state, while 6a-c have close to planar conformations, pointing to weak hydrogen bonds between the acidic C-Hs and the adjacent azine nitrogen atoms. The cytotoxicity of the six newly synthesized and three previously prepared compounds was assessed against a human glioblastoma multiforme cell line.

Resorcinarene-based cavitands with chiral amino acid substituents for chiral amine recognition

Resorcinarene-based deep cavitands alanine methyl resorcinarene acid (AMA), alanine undecyl resorcinarene acid (AUA) and glycine undecyl resorcinarene acid (GUA), which contain chiral amino acids, have been synthesized. The upper rim of the resorcinarene host is elongated with four identical substituents topped with alanine and glycine groups. The structures of the new resorcinarenes were elucidated by nuclear magnetic resonance (NMR), mass spectrometry (MS) and the sustained off-resonance irradiation collision induced dissociation (SORI-CID) technique in FTICR-MS. These studies revealed that eight water molecules associate to the cavitand, two for each alanine group. The alanine substituent groups are proposed to form a kite-like structure around the resorcinarene scaffold. The binding of AMA, AUA, and GUA with chiral R- and S-methyl benzyl amines was studied by 1H NMR titration, and compared to that of a binary l-tartaric acid and the monoacid phthalyl alanine (PA). The results show that these compounds interact with amine guests; however, with four carboxylic acid groups, they bind several amine molecules strongly while the binary l-tartaric acid only binds one amine guest strongly. The simple compound PA, which contains one carboxylic group, shows weak binding to the amines. The 1H NMR titration of AUA with primary, secondary, and tertiary chiral amines showed that it can discriminate between these three types of amines and showed chiral discrimination for chiral secondary amines.
