The synthesis of oxacalix[2]terphenylene[2]aromatics with enlarged macrocyclic holes by cyclooligomerization reaction of 5?-tert-butyl-(1, 1?:3?,1?-terphenyl)-3,3?-diol 1 with electron-deficient dihalogenated benzene and azaheterocycles is described. The structures of the macrocycles were characterized by NMR, HRMS spectroscopic and X-ray diffraction techniques. Single crystal X-ray analysis revealed that the terphenyl-3, 3?-dioxo unit incorporated in the oxacalix[4]aromatics scaffold can adopt all three possible conformations (I, II, III). The cis-conformational terphenyl-3,3?-dioxo (I and II) derived oxacalix[4]aromatics were found to adopt both chair and boat conformations, resulted in creation of molecular cavities capable of hosting solvent molecules of chloroform. The trans-conformational terphenyl-3,3?-dioxo (III) derived oxacalix[4]aromatics, however, adopt a twisted chair conformation with a narrow void space incapable of hosting any guest molecules.

Novel dyes based on the 12H-quinoxalino[2,3-b] [1,4]benzothiazine skeleton were synthesized and subsequently characterized using 1H NMR. Their electrochemical and spectral properties, such as absorption and emission spectra, quantum yield of fluorescence, and quantum yield of singlet oxygen formation, were measured. These compounds were evaluated as sensitizers for alkoxypyridinium salt photodecomposition, and the results are discussed on the basis of the free energy change for electron transfer from benzothiazine dyes to alkoxypyridinium compounds. Benzothiazine dyes are useful oxidizable sensitizers for N-alkoxypyridinium photoinitiators. The mechanism of the dye photobleaching is supported by time-dependent density functional theory (TD-DFT) calculations and the quantum yields of sensitized proton formation. Photoredox pairs consisting of benzothiazine dyes and alkoxypyridinium salt were found to be effective initiation systems for free radical polymerization of methyl acrylate and trimethylolpropane triacrylate (TMPTA) using visible light.

A direct and single-step method has been developed for the synthesis of mono and 2,3-disubstituted quinoxalines by using a AlCl3 induced (hetero)arylation of 2,3-dichloroquinoxaline. Both symmetrical and unsymmetrical 2,3-disubstituted quinoxalines can be prepared conveniently by using this method under appropriate reaction conditions. The reaction proceeds via C-C bond formation and can be utilized for the preparation of a variety of quinoxaline derivatives from readily available starting materials and reagents. The molecular structure of a representative compound was confirmed by single crystal X-ray diffraction study. Some of the compounds synthesized were tested for chorismate mutase inhibitory properties in vitro and one compound showed promising activity representing one of the few examples of chorismate mutase inhibition by a heteroarene based small molecule.

We report the synthesis, characterization, and cytotoxic and antimalarial activity of ferrocene-indole hybrids 8-14. The 2-phenylindole scaffold was chosen because of its potent antimitotic activity and ferrocene was chosen following the development of ferrocifens, ferrocene derivatives of tamoxifen, which are prototypes of a new family of organometallic anti-estrogens. Ferrocene-indole hybrids 8-14 and their corresponding organic analogues 1-7 showed only moderate antimalarial activities, while ferrocene-indole hybrids 11 and 12 showed excellent in vitro activities against the A549 human carcinoma cell line, with IC50 values of 5 and 7 muM respectively. These ferrocene-indole hybrids were up to 25-fold more potent as cytotoxic agents than their purely organic analogues.
